011 A new immunocompetent skin model recapitulates molecular features of psoriasis

Psoriasis is a chronic inflammatory skin disease involving interactions between keratinocytes and both the innate adaptive immune systems. The IL-17/IL-23 axis described to drive release of factors in (cytokines, antimicrobial peptides, chimiokines), which sit at heart an inflammation amplification loop. To this date, there are very few immunocompetent models recapitulating as close possible psoriatic skin. In context, aim work was develop 3D model lesional summarizing biological features pathology. As dialogue Th17 lymphocytes (LTh17) plays central role psoriasis pathogenesis, taking into account these two actors developed. A reconstructed epidermis (RE) co-cultured with differentiated LTh17. RE morphology well proliferation/differentiation imbalance response were explored. it reported lesions, results showed that IL-17 family cytokines increased. Histological analysis demonstrated mimic exhibited morphological changes. Moreover, mechanisms proliferation differentiation appeared profoundly altered while high keratinocyte characteristic observed. conclusion, we have succeeded designing vitro reproducing molecular alterations co-culture LTh17 led typical keratinocytes, thus proving successful communication compartments. This physiological relevant suitable for assessment natural therapeutic molecules dedicated treatment.
